We should be getting our 5 P800s on Friday afternoon. As it is the weekend I will people to take them home and play however I'm aware that the battery might need conditioning.
How long should I tell people to leave them on charge for before they start using them?

I won't pretend to be a battery expert, but my experience says that most manuals tell you to charge for 16 hours, however, since most modern devices have fairly intelligent charging circuits that switch off when the battery is fully charged, I've never really seen much point in it, I trusted mine to tell me when it was full

With the lithium battery of the phone, you only have to charge it until the phone reports it is charged. This should take anything between one and four hours, but as soon as it says it is charged, you can start using it, it wont damage the battery.

If you buy a new phone (with a battery), you should know that the phone, so also the battery has been empty for a while.
Maybe a few weeks in the shop, therefor a few weeks at the distributor, and therefor al long time at the factory.
So, the battery has been empty for a while .
So, I should advise that you charge that battery the first 4 times for about 10 hours.
But not only charging it for a long time, but also let it go
totally empty.
I think that that`s the best you can do

You can actually damage modern lithium batteries by letting them go totally empty, I'm being slightly pedantic, because the P800 (and most other devices) can't actually run it down completely, because it has high power requirements, so when it drops below a certain voltage, it just won't power up.
If however you were to connect the battery to a little bulb (or similar resistor) and run it till the light went off, the battery wouldn't ever work again, it requires a certain level of internal energy to maintain the chemical properties (or something along those lines).
As I said, I'm being pedantic, and since it will have been given a test charge at the factory, it'll still have plenty enough power to keep it healthy.
At the end of the day, just plug it in, new batteries only cost about $50, and you won't need one for a year or two regardless of how you charge it
